Antitumor , Antibacterial and Calcium Absorption Acceleration Effects of Chitosan Oligosaccharides Prepared by Using Ultrafiltration Membrane Enzyme Reactor

Abstract
In spite of various bio-functionalities of chitosan, the its effects in vivo were still ambiguous because of its low absorption on organism, Therefore, chitosan oligosaccharides(COSs) is necessary for an efficient utilization in vivo. COSs were prepared from chitosan using ultrafiltration membrane enzymatic reactor system and seperated three types of oligosaccharide, COS I, COS ¥± and COS ¥² dependent on their molecular weight and investigated their bioactivities such as antimicrobial activity, antitumor activity, and calcium absorption effect. Molecular weight distribution of COS I was 12,000 ~5,000 Da and that of COS ¥± 5,500~ 1,000 Da by Bio-Gel P-30 gel permeation chromatography. Main components of COS ¥² was trimer to heptamer(COS-3 ~COS-7: 600~ 1,400 Da). COSs were lower antimicrobial activities than chitosan. They, however, showed good activities for mainly pathogenic bacteria such as E. coli, E. coli O-157 and S. typhi in gram positive bacteria and Str. Mutans and St. ¥áureus in gram negative bacteria, and V. cholerae and V parahaemolyticus in vibrio bacteria from marine water. In antitumor activity of COSs for sarcoma 180 solid tumor cell, the activities of COS ¥± and COS ¥² were higher than that of COS I. Tumor growth inbition rate of COS ¥± wrere approximately 60% by every day injection of 10mg sample per kg mouse. In vivo using rats, that taken the diet added 1% COS to calcium chloride from oyster shell decreased about 75% of calcium content excreted from feces, and then, showed about 15% increasion in bone strength of femur. These results suggest that COSs could be an better biomaterial than chitosan for utilization in vivo.
